Peptide Reconstitution Math for Laboratory Protocols

Reconstitution is arithmetic, but it is the arithmetic most often done wrong when a vial label lists total mass rather than concentration.

The base calculation

Concentration equals vial mass divided by diluent volume. A 10 mg vial reconstituted with 2 mL of bacteriostatic water gives 5 mg/mL; the same vial with 1 mL gives 10 mg/mL.

Aliquot before freezing

Split reconstituted material into single-use aliquots so a protocol never depends on repeated freeze-thaw of one tube.

Record it

Log the lot number, diluent, volume, and date on the aliquot. That record is what lets a result be traced back to a COA months later.
